中文摘要 目的:探討耐力型運動與山苦瓜增補飲食對成長中雄性大鼠骨骼的影響。方法:將週齡7週大之實驗大鼠分為六組(每組n=12),依照飲食中山苦瓜凍乾粉末含量 (Mormodica charantia, MC) 與運動訓練 (exercise, EXE) 之有無,分成0%MC組、5%MC組、8%MC組、0%MC +EXE組、5%MC+EXE組、8%MC+EXE組等六組。耐力運動內容為22公尺/分鐘、60分鐘/天、5天/週之跑步機訓練,為期六週。實驗期結束進行犧牲,量測其體重與副睪脂肪組織重,並採集其血液、脛骨與股骨樣本進行各項分析;統計分析採2-way ANOVA ( = .05)。結果:經過六週不同山苦瓜劑量飲食與耐力運動後,運動 (p<0.05) 與山苦瓜飲食 (p<0.05) 分別顯著降低大鼠之體重,另外,副睪脂肪量 (p<0.05) 與瘦體素濃度 (p<0.05) 亦為接受運動訓練之大鼠顯著較低。海綿骨組織型態學分析顯示,正常飲食之大鼠(0%MCs)有顯著較高的骨量比率及較低的骨小樑間距,另外,骨小樑厚度之數值交互作用達顯著,事後比較發現山苦瓜飲食導致骨小樑變細,而運動可對抗此一山苦瓜之負面作用;在骨細胞活性參數方面,耐力運動與山苦瓜飲食分別對鹼性磷酸酶(骨生成)及破骨細胞數(骨吸收)有向下調節之效應。生物力學參數分析顯示,山苦瓜飲食與運動所引致之體重下降,使得一些與身體質量有關的力學參數在絕對值上(例如:降伏點負荷、斷裂負荷值及剛性)變差,運動訓練可部份保護山苦瓜飲食對骨組織剛性的負面作用。山苦瓜飲食之主要效果於CSMI (p<0.05)、橫斷面總面積 (p<0.05)、皮質骨面積 (p<0.05) 與皮質骨厚度 (p<0.05) 達顯著效果,山苦瓜飲食組顯著劣於正常飲食組,運動之主要效果僅於皮質骨面積 (p<0.05) 達顯著效果,運動組顯著低於非運動組。結論:六週山苦瓜飲食對各項骨組織參數呈現負面影響,運動介入可部份修正山苦瓜飲食對骨組織的負面影響。
英文摘要 Purpose: To investigate the effects of endurance exercise (EXE) and momordica charantia (MC) contained diets on bone qualities in growing rats. Methods: Seventy-two male SD rats (7 weeks old) were randomly divided into six groups, which were the 0%MC, 5%MC, 8%MC, 0%MC+EXE, 5%MC+EXE, and 8%MC+EXE groups (n=12 for each). After six-week experimental period, all animals were sacrificed. Blood and bone samples were collected, prepared and stored for various analyses. Two-way (MCEXE) ANOVA was used for statistical analysis (α=.05). Results: In exercise main effect, exercise animals showed significantly lower body weight (p<0.05), lower epididymis fat tissue weight (p<0.05) and serum leptin (p<0.05). In histomorphometry, the main effect analysis of MC showed that bone volume ratio and trabecular separation (p<0.05) of the rats feed with MC contained diets were significantly lower and higher, respectively, than the non-MC treated animals. In addition, interaction reached significant level in trabecular thickness that the 0%MC group was significant higher than the 5%MC and 8%MC groups while no difference was shown among three exercise groups; suggesting exercise’s efficacy in counteracting MC’s negative impact on trabecular bone. In bone cell analyses, MC and EXE respectively showed their effects in down-regulating bone formation and bone resorption. In biomechanical analyses, MC and EXE caused body weight decrement seemed to further reduced several size-related mechanical properties. Nevertheless, exercise somewhat modified MC’s related negative effects on bone. In tissue size measurements, MC fed animals showed lower cross-section area, cortical area and cortical thickness while exercise animals only showed lower values in cortical area. Conclusions: There seemed to be negative effects of the MC supplement diets on bone qualities. Endurance exercise showed some modifications on MC’s effects in bone material.
