The role of glutamatergic system in learning and memory has been extensively studied, and especially NMDA receptors have been implicated in different learning and memory processes. This study aimed to examine how the glutamate receptor ligands affected the cocaine-associated memory consolidation and extinction by utilizing the drug-induced conditioned place preference (CPP) paradigm. The mice were treated with D-cycloserine (partial NMDA glutamate receptor agonist), D-serine (NMDA glutamate receptor agonist), or sarcosine (glycine transportor I inhibitor) intraperitoneally at different time points to investigate the effects of the glutamate receptor ligands on the consolidation and extinction of the cocaine-associated memory. We have demonstrated that D-cycloserine (30 mg/kg), D-serine (300 mg/kg), and sarcosine (300 mg/kg) did not appear to affect consolidation of the cocaine (5 mg/kg)-induced CPP memory. In contrast, D-cycloserine (30 mg/kg), sarcosine (300 mg/kg), and D-serine (600 mg/kg) significantly accelerated the extinction of the acquired cocaine (10 mg/kg)-induced CPP memory.
