中文摘要 牛樟芝(Antrodia camphorata)為臺灣特有的大型真菌類,是相當著名且極為昂貴的藥用真菌,其子實體近幾年來有許多研究報告皆指出其具有多方面顯著的功效,包括抗氧化、保護肝臟、調節免疫、抗腫瘤細胞、抗發炎、預防心血管疾病以及降血壓等功能。本實驗目的是分析比較固體培養牛樟芝與野生牛樟芝其成分及生物活性。
已知大部分三萜類化合物為中極性化合物且具有共軛系統,在波長254 nm下具有吸收的特性,因此利用逆相高效液相層析法進行分析野生牛樟芝子實體、市售牛樟芝膠囊商品A和B、固體培養牛樟芝95%乙醇萃取物,從實驗結果得知市售牛樟芝膠囊商品A的確具有與野生牛樟芝子實體相似的中極性化合物;而固體培養基中含靈芝的量多寡並沒有與產生中極性化合物的含量呈現正相關;含固體培養基之牛樟芝粉末與沒含固體培養基之牛樟芝粉末知兩種商品, 均含有中極性化合物,但相對於野生牛樟芝子實體相當少。
在總多醣含量測定中,得知添加靈芝含量多寡並沒有與多醣體含量呈現正相關,固體培養基牛樟芝粉末(沒有加靈芝)具有較多的多醣體;在三萜類化合物含量測定中,含固體培養基之牛樟芝粉末與沒含固體培養基之牛樟芝粉末之兩種商品,其三萜類化合物之含量皆比野生牛樟芝少;麥角固醇定量得知,沒含固體培養基牛樟芝粉末並沒有比野生牛樟芝子實體較多含量;在總酚、總黃酮、縮合單寧含量測定中,趨勢則與清除DPPH自由基能力及還原能力互相呼應。
由實驗結果顯示:從HPLC圖譜中可以藉由分析固體培養基栽培之牛樟芝是否具有與野生牛樟芝子實體類似的三萜類化合物。藉由三萜類化合物定量可分析固體培養基栽培之牛樟芝是否具有豐富的三萜類化合物。在清除DPPH自由基能力及還原能力中與多酚類、黃酮類化合物、縮合單寧呈正相關,藉此研究之方法可檢測那種固體培養基栽培之牛樟芝具有較強之抗氧化能力。根據實驗結果,發現要人工培養牛樟芝與野生牛樟芝相同的成分是需要時間且模擬野生牛樟芝成長環境。
英文摘要 Antrodia camphorata(also known as Niuchangchih,AC)is an endemic species of macrofungi in Taiwan. In recent years, intensive studies has revealed that the fruiting body of Antrodia camphorata possesses significant biological activity, such as antioxidant, hepatoprotective, immuno-modulatory, anticancer, anti-inflammatory, anti-cardiovascular and anti-hypertensive activities. The objective of this study is to analyze the relationship between the ingredients and bioactivity for the solid media cultured and wild-type AC.
It is known that most triterpenoids show medium polarity and with the conjugated characteristic, which has an strong absorption at 254 nm. So, this study employs the reverse-phase high-performance liquid chromatrography to analyze the 95% ethanolic extract of wild-type AC, AC merchandise A and B in market, and solid media cultured AC. The results show that merchandise A contained ingredients with medium polarity as that of wild-type AC. No matter the solid media containing how much of the Ganoderma lucidum, the AC did not show proportion relationship with the production of medium polarity compounds.
In the determination of total polysaccharide content, the result shows that the addition of amount of Ganoderma lucidum in the media does not show positive correlation with the amount of polysaccharide. In the determination of triterpenoid content, it was found that natural AC contains more triterpenoids than solid media cultured AC. In the determination of ergosterol contents, it was found that natural AC contains more ergosterols than solid media cultured AC. In the determination of polyphenol, flavonoid, and condensed tannin contents, the trends match the DPPH radical scavenging activity and reducing power.
The results reveal that we can analyze solid media cultured AC from HPLC profile to determine the triterpenoid content. The DPPH radical scavenging activity and reducing power are positively correlated with the amount of polyphenol, flavonoid, and condensed tannin, so the way used in this study can be used to analyze whether solid media cultured AC contains antioxidant activity. Based on this study, it was found that to cultivate the AC with the comparative components with nature AC needs time and mimic environment.
