中文摘要 本論文主要探討利用固態或液體浸漬培養產製食藥用菇類之生理活性及產菇效率。以金鼎側耳及阿魏菇分別進行固態及液態浸漬發酵,取得子實體、菌絲及發酵液,以動物及試管方式進行不同生理活性,包括抗抗氧化、抗致基因毒性、抗高血糖及抗高血脂活性探討;並且亦進行菇類固態栽培研究,將猴頭菇培養於使用農產廢棄物部分取代木屑之培養基中,探討出菇之生物效率。另外金鼎側耳培養於不同環境因子,包括二氧化碳濃度及不同波長光照,探討出菇品質。
結果顯示,阿魏菇之六種萃取物中,EAM (溶劑萃取物)具有最高DPPH 自由基清除能力及鐵還原力。並且,在抗各種試驗的突變劑時,EAM有最高的抑制率,亦有最高的抗DNA損害活性。
金鼎側耳的抗氧化及抗高血脂活性試驗,兩種活性之間可能有關聯,而兩種試驗樣本 (EAE及MOE) ,在動物及試管試驗中,具有最高活性。且於動物試驗中,此兩種樣本之動物試驗組,其肝細胞液泡化程度較其他試驗組為少。
由金鼎側耳發酵液萃出之水溶性多醣類具有抗高血糖效果。取其多糖類餵飼糖尿病老鼠,體重減少量較不餵飼多糖類者為小。禁食血糖量較對照組低44%。此情況亦呼應早先有關多醣體之抗人工轉移肺腫瘤及免疫促進效果之研究結果。
在固態發酵中,兩種猴頭菇使用農產廢棄物作為栽培基質是可行的,且最適的培養條件是23℃、90% ERH及含500 g/kg蔗渣之培養基有最高的產率。另外,調控栽培環境的二氧化碳濃度及不同波長光照,栽培金鼎側耳,結果0.1%–0.15%的二氧化碳濃度及波長為720 nm者有較高的出菇品質。
綜合以上結果顯示,由試驗的菇類所得之萃取物具有一些優良的生物活性功能,包括抗氧化、抗致基因毒性、抗高血糖及抗高血脂;並且藉由調控栽培因子將有助於固態栽培菇類之出菇品質。
英文摘要 The research is mainly aimed at physiological effects and fruiting efficiency of edible and medicinal mushrooms with solid-state or liquid submerged culture. The extracts of fruiting body, mycelium, and fermented broth from Pleurotus citrinopileatus and P. ferulae with solid-state and liquid submerged culture, respectively, were applied to study the physiological activities, including antioxidant, antigenotoxic, antihyperglycemic, and antihyperlipidemic effects in the in vivo and in vitro. Moreover, in the solid-state cultivation of mushroom, the agrowastes partially replaced components of substrates to cultivate two Hericium spp. for the exploration of biological efficiency. The P. citrinopileatus was cultivated under controlling environmental conditions, including concentration of CO2 and illumination of different wavelength of light, for studying fruiting quality.
The results showed EAM (a solvent extract) have the highest DPPH free radical-scavenging activity and ferric-reducing ability among six test samples in the study on the antioxidant and antigenotoxic activities of P. ferulae. Meanwhile, the EAM exhibited the highest inhibition rate against every tested mutagen at a low concentration of 0.08 mg/mL and a high concentration of 0.3 mg/mL in each plate, and it had also the highest anti-DNA-damaging activity among all test samples.
There may be an important relationship between the antioxidant and antihyperlipidemic activities of P. citrinopileatus, and the higher antioxidant and antihyperlipidemic activities of Pleurotus citrinopileatus both in vitro and in vivo, were found with two test samples (EAE and MOE). Hepatic cells were less vacuolized in the MOE- and EAE-groups than in the other experimental groups.
The water-soluble exopolysaccharide from fermented broth of P. citrinopileatus has an antihyperglycemic effect. The diabetic rats fed with water-soluble polysaccharide of P. citrinopileatus lost less body weight than those fed SPPC-free regular diet. The fasting blood glucose levels of diabetic rats fed with SPPC were 44% lower than the negative controls. This corroborates the findings of previous studies in the anti-artificial pulmonary metastastic tumor and immunoenhancing effects.
In the solid-state fermentation, it displayed high in cultivation of H. erinaceus and H. laciniatum and indicated that the substitution might be a feasible proposition. The optimum conditions for reaching the highest biological efficiency and yield of the two Hericium spp., were at 23℃ and 90% ERH by using 500 g/kg of sugarcane bagasse to replace sawdust. Besides, the optimum wavelength of light illumination and atmosphere in the incubator might benefit the yield and quality of the mature fruiting body of P. citrinopileatus. A higher fruiting quality should be obtained with an atmospheric CO2 concentration of 0.1%–0.15% and a wavelength of light of 720 nm.
In conclusion, the results suggest that the extracts from test mushrooms have fair bioactive functions, including anti-oxidant, antigenoxicity, anti-hyperglycemia, and anti-hyperlipidemia. Besides, it is contributive to fruiting quality via regulating solid-state culture conditions.
