中文摘要 重力(gravity)對生物系統的影響隨時存在且深遠,過去鮮為人探討;然而隨著近年人類頻繁的太空探索及相關生命科學的研究,重力對地球生物的影響漸受重視。重力加速度高於一個地心引力即可稱為高重力,現實環境中人類在某些特定的情況(如:乘坐雲霄飛車,飛行高地心引力G動作)均會承受高重力。人體暴露於高重力環境時,身體各系統皆受到高重力的影響,對腎臟泌尿系統的影響可反映在尿液參數的改變上。利用人體離心機產生之高重力,研究受測者暴露高重力環境前、後尿液性質的改變並結合分子生物學的研究,分析高重力環境下尿液微小核糖核酸(microRNA)變化。高重力可導致人類尿液結晶增加,尿液參數及微小核糖核酸表現的改變。這些結果顯示高重力環境對人類泌尿腎臟系統有特定的影響,如此一來可以開拓高重力在生物醫學領域研究及臨床醫學運用的潛能。
英文摘要 Hypergravity effects on human are tremendous. However, there are not enough systemic studies on renal and urology systems. We used human centrifuge to generate acceleration (hypergravity) on human subjects for study. The impact of hypergravity on human renal system can be directly reflected on the changes of urine properties. The urine samples before and after the centrifuge ride were collected and analyzed. Urinalysis for urine properties (specific gravity, pH, urobilinogen, protein and crystal) and expression of certain urinary microRNA (miR-21, -151, -210 and -18a) were investigated. We explored the potential role of human centrifuge riding as a research mode for acute kidney stress and nephrolithiasis in human. Change of urinary microRNA expression from this study could be served as a potential biomarker for acute kidney stress.
