中文摘要 幽門桿菌為引發胃腸潰瘍的胃部重要致病原,並且被 WHO 認定為胃癌之第一型致癌物。幽門桿菌感染的盛行率於糖尿病病人是增加的,治療成功率相對較無糖尿病者為低,且於殺菌成功後容易再感染。因此,評估幽門桿菌的毒力於高葡萄糖濃度下是否被加強具有其重要性。以幽門桿菌標準菌株 J99、ACTT43504進行體外實驗分析。本研究測試在不同葡萄糖濃度 (100, 150, 200 mg/dL)之下,是否可增加 (一)幽門桿菌的生長; (二)幽門桿菌對於胃癌細胞株 AGS 的附著能力; (三)毒力因子 CagA 的表現量。由結果顯示在 200 mg/dL 的葡萄糖濃度之下,兩菌株在48小時達到最多菌落量,且維持至72小時; 在200 mg/dL 的葡萄糖濃度之下,細菌的附著能力明顯優於100mg/dL 及150mg/dL 的葡萄糖濃度之下; 而細菌毒力因子CagA 蛋白及磷酸化CagA 蛋白 (p-CagA) 的表現也是在較高的葡萄糖濃度(200mg/dL)明顯增加。研究發現,高葡萄糖濃度可以維持幽門桿菌的生長,增加附著能力及CagA/p-CagA的表現量。
英文摘要 Hyperglycemia increases the risk of gastric cancer in H. pylori-infected patients. These suggest high glucose may affect H. pylori or its infected status. Two strains of H. pylori were used to investigate whether H. pylori growth, viability, adhesion and CagA-phosphorylation level in the infected-AGS cells were influenced by glucose concentration (100, 150, and 200 mg/dL).The results showed that the growth curves of both strains in 200 mg/dL of glucose were maintained at the highest optimal density after 48 h and the best viability of both strains were retained in the same glucose condition at 72 h. Furthermore, adhesion enhancement of H. pylori was significantly higher in 200 mg/dL of glucose as compared to that in 100 and 150 mg/dL (p < 0.05). The cell-associated CagA and phosphorylated-CagA was significantly increased in 150 and 200 mg/dL of glucose concentrations as compared to that of 100 mg/dL (p < 0.05), which were found to be dose-dependent.
In conclusion, higher glucose could maintain H. pylori growth and viability after 48 h. H. pylori adhesion and CagA increased to further facilitate the enhancement of cell-associated CagA and phosphorylated CagA in higher glucose conditions.
